Best app/site for usability and accessability testing

Hi all,
I just wondered what other developers/agencies use to test sites prior to launch.. Does anyone recommend any apps or websites in particular?

I realise that a lot of testing will need to be done by humans but are just looking for something that will save us time & energy to make sure sites pass general usability and accessability tests as well as general markup.

Any suggestions appreciated!

Cheers Pho. Will probably be implementing a CMS for the school I work at so stuff like this could be really useful to me.
