My next suggestion is one your going to hate, but fortunately doesn't cost you a penny!!!

Build the basic parts of the pc completely outside the case, place the motherboard onto a piece of cardboard.

I have a laptop that has bad "squealing" and hissing interference that is even more pronounced when you are doing something, this is when using the headphone jack, as soon as I use a USB device for my audio this sound goes away.

From what I have read this is just down to bad quality on-board sound and the cables picking up interference along the way, it is worse in laptops as things have to be more compact and closer.

I would say it was something else then. How much of your PC do you want to take apart? lol

For me the next step would be the audio cable, but I would move the rad rather than the cable and see if it still does it then.

ohh god this is getting carried away....
this means i'm going to have to dismount fans to get to the mobo screws, and i've nearly annihilated the cross on the head of the top fan screws, making it tricky to do anything with them.
If this is the only remaining option, I guess i don't have much choice, but if there are ANY other ideas at all, now would be perfectly perfect to voice them.
dandos i've already disconnected the front audio altogether, so its completely out of the equation.

also, just googled a couple of the chip names, turns out that the major audio chip is in the bottom left of the motherboard, (basically under where i JUST moved my GPU to, between the last two PCI sockets). Problem is my case is small, meaning this is closer to the power supply than Asus probably intended.
Potential issue?

goddarnit

not entirely prepared to strip out the entire thing, but if it IS the power supply causing fusses (see my previous post), scooping that outside of the case while leaving the rest in might be an alternate approach

you need to short the two pins that the case switch connect to (only for a short time though), you could use a screwdriver, if you are not comfortable doing that you can just hook up your case switch to it.

Without putting everything back in the case just yet, can you connect the audio cable to the motherboard and check that there is no interference there as well?

What CPU cooler are you using with it out of the case? Is it still the H50? If so try just holding the rad of it near where it will be while it is in the case and see if that creates any interference, if that does not the do the same with the PSU as well.
