So it looks like I'm going to go with a P600s case and an Arctic Freezer II 360 AIO... looks like it will fit snug at the top.

Case appears to come with 2 front fans and one rear, question is, is this likely to be enough or do I need to replace/add to those fans?

12900k & 3080ti FE will be in the box, on probably an MSI Z690 Carbon Wifi.
 
How do you plan to set it up? Top EXHAUST for the AIO? I Google'd that case and it seems to have solid panels that are removable and show mesh. The front looks somewhat restrictive, so some Arctic P14s might be a good shout there, if you keep that front panel on. I used those on a Fractal Define 7, which is pretty restrictive with the door closed, but it worked absolutely great in that scenario with the door dampening noise and the high static pressure of those fans able to pull air in through the side slats and then the filter.
 
Despite the case allowing triple 120mm fans on the front, 2 pre installed 140mm fans should be a better option, anyway. Can’t think of any possible improvement, to be fair.
Cases which allow meaningful bottom intake may help greatly the GPU, but not an option for the P600s.
I would use the top AIO as exhaust.
 
The 3080ti will dump a lot of heat inside the case, for sure, but even with that heat going to the AIO, the arctic should still be plenty for your CPU, at least if you don’t plan to go nuts overclocking it.
The only thing I can think about is, if you try change the rear exhaust as intake, and monitor if improves anything, but nothing else would make sense to change.
I recommend HWinfo to check temps. I would keep notes of GPU core and GPU memory junction, NVME temperature (make sure the hot air from the GPU doesn’t push your NVME to thermal throttling, and CPU temperature, for sure. MOSFET should be gold with the included fan on the Arctic Freezer. Not a gimmick.

I have just done a build in the P600S (good choice, it's an utterly superb case, best I have used), and the fans that come with case are pretty good, and quiet (I only replaced them for aesthetic reasons), you'll have no problems.

As good as stock 140mm case fans are. 2x 140 will supply air 360 rad's 3x 120mm fans use. That leave your 3080ti FE w/ 3x 80mm (guessing) needing at least 1 more 140mm intake. Moving rear exhaust 140mm to front will help and hopefully be enough for both rad and GPU. I would definitely keep the front center vent cover removed to give fans as much unobstructed vent area as possible. Try running with no rear exhaust fan. Most of my build don't use any exhaust fans, but with 360 top rad a rear fan may help move GPU heat out back instead of it flowing up and out thru radiator.

If stock case fans can't move enough air, cheapest higher airflow option is Arctic P14s which are not bad. While not great, they are a good performing low cost fan. There are better 140mm fans but they are also more money.

Had a right old laugh getting ram delivered but sorted now. Case is fantastic, definately worth the money over a cheaper one.

So far so good. Cinebench hits 86 degrees max on multi core and I've not had any problems with the 3080ti so far.

Very happy with it all.
