I have an MSI R9 290x, connected to a LG 34UM95 by Displayport cable. I can't get any audio over Displayport, I don't seem to even get the option in the Windows (8.1) sound panel. I think I'm using onboard sound on my MSI Gaming 7 Z97 mobo; should this work?

It should. Things to check: All drivers are installed and up to date.
And have a look in "playback devices" under Windows (Right click on the speak bar down the bottom right of the task bar). You may need to change the default

This what I see in the Sound properties:

If I click properties on the top one, it says HDMI jack. I don't see anything referring to Displayport.

Should I be able to be use onboard sound and have it output via DisplayPort?

What if you scroll down the list does it display anything else that it sees as "connected?"

Another thing is to right click on any of the devices and click on "show disabled devices" just in case some software has set it to disabled.
