Can update my bios with my PC fully built and windows installed on it

Can you? Yes, no problem. Always back up windows and files to 2 different sources should issues arise, but this is extremely unlikely to be required. Check your motherboard manual, and make 100% sure you have the correct bios. Follow the instructions perfectly.
Bios updates are easy and quick, but can nuke some boards if done wrongly.

You can't? Can you be more precise with where the issues lays.

Never use Windows softwares for updating BIOS, unless liking unnecessary risks.
Most boards have had very easy (and least risky) update tool inside BIOS itself for nearly 20 years.

Be aware that any settings you have made to your bios (overclock for example) will be lost when you update.

Write down or take photos of any changes you have made so you can input them once you have updated the bios.

If it is a prebuilt computer, may be best asking the vendor if it will affect the warranty in any way.

Yes you can and it’s very easy. Download new bios, save file to usb stick, boot to bios, flash bios from there.

Bear in mind that it’s not without risk. If you have a power cut or similar mid update you can end up with a bricked board.
