Since I got very poorly referenced alphacools's pump/res combo for my first wc build, I ended up connecting pump's out to cpu block's out. Only realised this when the water started flowing. Is there a difference in cpu block's in and out? Should I redo my tubing?

If your cpu block is labeled In/Out then yes otherwise it won't be working properly. Modern cpu blocks are designed to only work one way.

I've done this and feel your pain. As said, change it if the block is marked in/out

Difference yes, but if it makes any difference to you we're talking "temps not as great as could be" difference, not "omg it's break stuff!" difference.

I just checked my EK Supremacy Acetal and it doesnt have the IN/OUT markings....

The hole closest to the centre is the IN, the one furthest away is the OUT (would prob have said in the instructions).
