I was wondering;

What is the difference between the Gigabyte GA-EP43-DS3 and the GA-EP43-DS3L?

Nope. Both support DDR2 so I did a comparison from the gigabyte site. Both got the same chipset. Some difference in PCI slots. Same storage interface bits. The DS3 has loads of firewire options. USB stuff is the same. The DS3 has an internal I/O connector for firewire. DS3 carries a back panel firewire connector.

So the DS3 if you like/need firewire, DS3L if you don't?

Same as DS3 and DS3P just had different capacitors.

Yes, L means just Lite version
