It's just Asus rerouting PCIe lanes from the usual bottom expansion slot to somewhere with more room.
Had a look at the manual but it doesn't state if it's using lanes from the CPU or chipset. For the M.2 slots already on the board, the top is CPU controlled while the bottom is chipset controlled. I'm going to take a wild guess that seeing both x16 slots and the top M.2 slot uses all 20 CPU lanes, that the DIMM.2 slot is chipset controlled, therefore I would be tempted to use the top M.2 slot for the OS already there.
