Thinking of establishing a point to point link between two locations approximately 400m apart. I believe they have line of sight, so would a pair of directional wi-fi antennae (think yagi is the type we need?) be able to work across that distance?

Yagi will most likely work, i've had a 22dBi dish antenna over a distance of 1.2 miles before and that worked well enough to stream (fairly low res) video.

The access points were 11meg and pretty domestic (USR and a Linksys i think it was)

I have a 18Dbi Yagi and can get to an AP a mile away (Nothing special at the other end, the AP is even in a building through a wall) at 2Mb/s (downloading at approx 40KB/s)

Putting my own equipment at the other end (even just an access point with the antenna outdoors (nothing special) would give me much better results, and a directional antenna at each end would be ideal for higher bandwidth applications.

I found a UK retailer of all the outdoor antenna's / pigtails / connectors / cables needed for wifi.
They are in an industrial unit in the middle of nowhere, and their online live chat is great (in building) so you can just ask them if they have x,y and z in before you turn up, they also deliver. (was very impressed, can you tell :P?)

I got this from them:

Which coupled with my Ubiquiti SRC PCMCIA card (pictured) is awsome!

Nice one. Looks like I might be able to get 10mbit+ up and down in that case. (I'm going to pay for an antenna on the roof of a datacentre...)

You should not expect to to get high speed both up and down at the same time, the connection has to share the total link capacity.

How symetric is the data exchange? all up, all down or bit of both?

Those distances, perfect line of sight using 802.11g I would say 24Mb/s would be an easily reachable goal with good kit at each end, giving you 10Mb/s each way without a problem but errata is right, you must take into account both upload and download.
