Do these actually work?

The first one might in the right situation, with the right hardware and the right software. But note Microsoft and third parties don't do validation testing with these settings on anything but their default assigned values. So, potentially, you'll be harming stability.

The second one is only to be used by driver developers. Period.

I love how they just pluck the "if you have 512MB or more memory" figures out of thin air. Microsoft doesn't really document these settings because they are for internal use only. So it really makes you wonder where these tweak guides get their information. They make it up. Not a good thing to do when the author doesn't really understand the forces at play.
