I know these questions are subjective and asked many times but I am aware there are serial upgraders here and hope one of you may have experienced the upgrade.

I have an E8600 100% stable at 4.3GHz. It's been perfect for over 4 months now, having previously sat at 4GHz for over 2 years. I consider my PC to be quick. Is there any good reason why I should go for an 2500K or higher? The PC would need to be at least half as quick again before I changed up.

I can actually answer this with actual end user experience :)

I had an E8500 @ 4Ghz, 4GB DDR2, HD 6870 rig before the one in my signature so fairly similarly specced to your current one.

I then upgraded my motherboard, CPU and RAM only and kept all of my old bits. The difference was really quite large in games that I play. I couldn't give you a 50% upgrade confirmation as I didn't benchmark or measure before I ugpraded but for what I paid it was massive.
 
A lot would depend on how you use your pc?

A. Is it just web browsing, light gaming etc....
B. Or do you encode videos, heavy gaming etc...

If its A, then i dont think you will see much difference.
If its B, then as Rusty said, you should see a decent difference in performance.

I am still on a Q6600 @ 3.4ghz, 4 GB ram and a HD 5850, i do play a lot of games, and it is starting to show its age, but it can still run most games on high settings with a decent fps.
 
Just something to consider, the 2nd hand value of your cpu and mobo is actually pretty decent and if you don't mind buying 2nd hand parts for your upgrade you could effectively upgrade for free. Guide prices for parts I've bought recently:

i7 920: ~£75
X58 mobo: ~£65
8-12GB DDR3: ~£25

I know X58 is a few years old now but considering how cheap it's getting it's actually pretty good value for money imo. Otherwise if you spend a little more on the chip you could get a 2500k and P67/Z68 setup for only slightly more.
