I wouldn't buy the Wii version purely because i resent them forcing you to insert the Wii remote into the guitar. While this isn't hassle, you have to consider that it means the wireless adapter isn't built into the Wii guitar yet it costs EXACTLY THE SAME as the PS3 and 360 guitars that have it installed.
DLC is downloadable content. It means that new songs get released via the Playstation Network (or Xbox Live if you have a 360) and you can pay a small fee (couple of quid) to download them.
It means once you've run out of songs to play, you are bound to find something on PSN/Live that you like and you can download it
Occasionally there are free track packs as well for download.
