Which format is best to use to create a HD video?

I've seen a lot of HD files that are MKV - what are these?
They're approx 4-5gb for 720p, is that about right (edit: for a film)?

MKV (Matroska) is the most modern video/audio format around. Well, as Chris says, it's container rather than a format because they can contain differing formats/codecs.
x264 being the most popular encode for HD
(Hope I make sense!)
