Is there much of a real noticable performance difference between the E4400, E6400, E6420 and E6600 when all run at 3.0GHz?

Nope

Depends what you do

but I doubt if you had 4 pc lined up and you used one after the other you could go...

Yeah I would bet on it. assuming no synthetic benchmarks were run :p
 
From the benchmarks I've seen the 2xxx are about 5-10% slower than the 4xxx which are 5-10% slower than the 6xxx.

Can't remember the link offhand but someone posted it in the 2xxx overclocking thread.
