Well, after planning my loop in my head and ending up with a few wrong estimates. I had to change a few tubing routes. Only to realise I forgot to re adjust my CPU block.

As of now, the water is coming in at the OUT port and coming out at the IN port.
How much difference is this going to make if any?
If it helps, I have my CPU depidded and direct die mounted with CLP.
 
massive difference, the block will have a jet plate innwhich is supposed to speed up the flow of fluid over the block itself, putting it in the wrong way round will mean it is doing the exact opposite and massively slowing flow over the block
 
Figured as much regarding the jet plate.
Although I thought it would only be 2-3C.
Guess I'll need to find a way to drain it enough that no water will come out of the tube/rad when I take the tubing out.

Good luck.... I had an issue when rebuilding my loop recently where one of the fittings onto my CPU block was leaking. Ended up having to drain the whole system down and still ended up wrapping everything in towels. Every time you think enough water is out of the system and you can start taking tubes off more water appears from nowhere... I just ended grabbing some bath towels and jamming them round everything. made me very popular with the missus as I'm using blue coolant that stained :-)
