MBP Spring Cleaning - how?

Had my MacBook Pro 2011 for about a year. It was brand new, just unopened for 6 months. Anyways I've been working pretty heavily on it, mainly video editing so there's a lot of footage and associated shiz laying about on there.

Anyways recently I've noticed it's running like absolute chode, so I'm looking todo some sort of spring cleaning on it. I can't imagine that having lots of video files on it is causing he slow down as they're not in use, just sitting idly.

What steps can I take, programs to use etc, that will help speed up the system again?

Onyx (free), or CleanMyMac (paid)

Something like GrandPerspective (free), DaisyDisk (paid) lets you visually see what and where is taking up the most space.

Out of habit I also regularly run Disk utility > Repair Permissions

What I use to. Must admit when I got my first macbook I did the Repair Permissions a fair amount. Can't think I have ever done it on my new one.

Cleanmymac is essentially one click to free up some disk space whereas Onyx is much more powerful if you know what you're doing.

I always repair permissions out of habit from what I've read and it tells me it found errors and fixed them, but unsure as to whether it actually makes any real world difference?
