No, but if your PSU has separate rails then make sure you don't overload one. (though I think the molexs usually have their own anyway so it shouldn't matter)
 
ive used one. as long as you dont overload one rail it should be fine. i mean as long as your not trying to run 2 gtx of one 12v rail with molex. generally though i recommend using the pcie connector from psu.
