Looking at speeding up the system a little, especially when the gf is running multiple applications for her uni work. Its currently running 4gb 667mhz ddr2. I'm thinking of either faster 8gb ddr2 at 1066mhz or 4gb ddr3. Not sure which would be more suitable. I mainly use it for photo stuff. It also supports a media extender box.
The rest is:
Asus P5kc P35 so will take either 4 slots of ddr2 or 2 ddr3
Vista 64, though soon to be 7
Quad 6600, stock speeds, though could be clocked i guess, its got a silent knight cooler
Hdd will be upgraded to 1tb samsungs in raid in near future.

Any thoughts/advice appreciated, i'm started to get confused

With 4GB of DDR2, I wouldn't have thought the RAM will be limiting performance (unless you are running into memory limits).

I think its more likely that the 2.4GHz CPU is holding you back.

I suggest giving the CPU a quick overclock, 3Ghz should not be difficult at all. If you feel more confident, you can try clocking it to the max, it will give you a nice performance boost.

I agree with the above here, a quick overclock which should be pretty easy to acheive to 3.0ghz which will also speed up the ram by overclocking it to keep up with the processor.

You said you mainly use it for photo stuff? Are you dealing with particularly large files , in which case is the memory being eaten up when doing multitasking aswell as handling lots of photos?

If thats the case then the memory might be a limiting factor in which case moving up to 8gb could be seen as wise, and if its a decent ram you would still be able to achieve some overclocking with the processor (albeit being limited by the amount of ram here)
