When using a radiator vertically, does it matter whether the ports are at the top or at the bottom of the rad?

It is a Alphacool Nexxxos XT45 120mm

yes it does, air will lock at the top of a rad if the barbs are at the bottom. To fix that uou have to rotate the case untill all air is out.

Another question then,

My rad has three ports in both intake and outlet. With the ports uppermost, would it be a good idea to fit G1/4" central heating radiator bleed valve(s) to the upper facing port(s)?

The radiator top will be the highest point of the system. To bleed the system then all I would need to do is temporarily remove the DVD drive.

Alternatively one could be a fill port.

I would look to bleed the system by rotating the case gently until all the air is out. Once the air is removed, there would be no use for the bleed ports that you mention.

Actually, if you've got upper facing ports on the highest point in the loop, you could simply use that as a fill port, good idea.

It doesnt matter as such but you will get some trapped air at the top but can be removed with turning the case upside down, their wont be no noticble difference in flow if the ports are at the bottom

I've put 2x280 rads on top of the case, free to move, so whenever i fill my system i just rotate rads so that end ports point up and all the air in the system goes straight into the res. Not for everybody. If you have mounted your rads in the case i'd say a fill port at the top most part of the loop would be a good idea. Also it depends on the order of the loop, my rads empty into res/pump combo, so all the air doesnt have to travel through the system to get to reservoir...
