My brothers PC was working fine this morning, he shut it down however now it just won't turn on at all. Pressing the power button over and over and nothing happens.

Take the side of the case off, everything is connected fine. Nothing is lose. All of the case buttons are connected correctly to the mobo.

Specs:
E6600
4GB DDR2
8800GTS
Hiper 550W PSU

What is the safest and easiest way to test a PSU?

Are you able to change the psu cable? Could be lucky and only fuse has gone.

Are any lights showing up on the motherboard ? Even when pc is shut down motherboard lights tend to still show up.
 
Try using a paper clip on the 24-pin PSU plug and short the Green to any Black cable, and try a power on, if the PSU is working, then the PSU fan, and any other fans connect to the PSU should spin up
