Short and to the point-

Say I have a shape in one layer and I want to cut that shape out of another layer, How would I do that?

CTRL+click the actual layer that you want to copy, that will create the selection of the shape for you.

Go to your new layer, Select > Inverse (if necessary?), and click the Layer Mask icon.
