Right got my loop all tubed and clipped up. And I know how to power the pump using the PSU and not powering rest of PC, but when should I put the coolant in before or after I power the pump?

Before.

You should never be running the pump dry as this can damage it. When you start up the pump for the first time, you'll probably need to keep stopping it to top the loop up as it drains of air as well.

Yeah, get as much fluid into the system as you can before you start the pump, then start it and keep filling the res up as it pumps round.
