Over the last few days I've just been trying to work out how to upgrade, may sound stupid but I'm just not having any luck, really hope someone can help me out here because I have no idea what to do.

I have bought a Windows 8 Pro key from Microsoft, I go into Windows 8 Upgrade Assistant, I click "Install now". I get this. :

Okay, I try to install with USB. Doesn't work, I can boot from it, but then it tells me I have some missing drivers (doesn't say what ones) and will not continue with installation, have to go back to Windows 7.

Then I try the last option: Making a ISO file... You guessed it, doesn't work either. I get THIS error message:

Does anybody have any suggestions? I'm at a loss here. I've tried rebooting, re-downloading the upgrade assistant from various sites, nothing has worked so far.

I think the fact that you can boot from the created USB stick speaks volumes. There is clearly a driver (most likely storage) missing from your machine during setup (did you need to use your motherboard's RAID/AHCI drivers during Windows 7 setup?) and as such the installer cannot proceed.

However just to rule out a corrupt download (has happened to me once), delete the hidden folder C:\esd, then start the whole process again, which will obviously re-download the files. Make sure you have no third party firewalls, or more importantly no proxies (my first problem was in the office trying to get out to the internet using Forefront TMG).

If you are continuing to have problems after that, drop me an email.

What hardware are you trying to install onto? have you checked that all the core hardware chipsets graphics and storage controllers are supported? sounds like you need to find some win8 drives to load on boot to me.
