Depends on the OS you are using. Win 7 was able to do it, but it had its failings. Windows 8.1 is far superior in this respect, but there is no guarantee!
 
Just be sure to remove the chipset and drivers for the P8H61-M LX and then install all the P8H61-I R2.0 drivers, as they will be different.
I've done this a few times and never had any problems. You may have to re-activate Windows, depending on whether you have retail or OEM.
 
Personally if your changing motherboards I always reinstall windows, as it give s much cleaner and faster machine (compared to trying to remove all the old drivers and then installing new ones).
 
You're going from an H61 board to an H61 board - you'll be fine. Just uninstall motherboard-specific drivers like LAN and sound before you shut down.

I've gone from a P35 board to a P67 and Windows 7 didn't even blink.
 
I did a P55 to H61 motherboard and CPU swap yesterday without reinstalling Windows 7 and it all worked fine. Takes a bit of time installing all the new drivers and getting rid of the old ones but no major hassle. Try first without a reinstall. You can always do one later if Windows starts misbehaving.
