I have my sky Q mini box connected to a 4k TV via HDMI. I have my PC connected to the same TV via HDMI and also to an X34a via DP cable. I have all PC content coming through my PC headphones, but obviously the Sky content is coming through the TV speakers but I want that to come through my headphones also.

Could I run a digital audio cable between Sky box and PC? Or, would I have to run the DO cable from the TV to the PC?

You'd need a sound card with optical input to run a digital cable from the Sky box to PC. Plenty have optical out, just a motherboards do, but they are output only. Sound blaster Z is possibly the only mid budget card or under that has optical input.

You could use an analogue cable to the line in on your motherboard. Note sure if there is any analogue output on a Q mini box. If not you'd probably need to connect a cheap DAC to optical out, then run an analogue cable from that into the line on in the PC motherboard.

No analog outputs on a Sky Q mini, HDMI and optical only.

I'd look for a cheap stereo amp that accepts optical and has headphone out.
