SQL Server - Slow and Unresponsive

We have an Intel Xeon 2.8GHz Dual-Quad Server, it's basically one of our SQL 2005 Servers. I am having issues with it running slow and unresponsive. The CPU usage chart in Task Manager is constantly going up and down however under the processes tab - it indicates it's mostly idle. Paging issue? I have attached a screendump of Task Manager below. The process "mssearch.exe" has an extremely high "I/O Reads" count, is this normal?



On this server alone, we have several databases but only one which is accessed constantly. Its size is ~70GB with ~250 users. Where's the best place to look for problems, performance seems to be deteriorating as data grows.
 
Is it the server's response that's slow, or just SQL? Might help if you narrow it down - there could be loads of causes!
