Our current system has now outgrown our DDS5 / DAT72 backup system so I am looking at options to replace it.

DAT160 is out as it only offers 80gb native and DAT320 is too expensive compared to SDLT320, AIT and RDX are out due to the cost of the media.

This leaves LTO and SDLT, I am in the process of deciding between LTO2 and SDLT320 and am hoping people can give me some opinions on which is best as they seem pretty evenly matched.

Both do the job - I'd look at the software you want to use and then check compatibility. I have used both in the past and noticed, normally, that LTO could handle more tapes for the same price as SDLT. But things may have changed since then as it was a while ago.

Go with LTO, stay away from SDLT if you can.
LTO is far more widely used and supported and the hardware also tends to be much better.
Maybe try and go upto LTO-3 if budget allows to give yourself some future growing room, LTO-2 is getting a tad old now.
 
Seen as though you're currently using DAT72's, a LTO2 drive will suffice, as I'm assuming seen as though you're probably using a 2 tape per day - equalling ~140Gb, LTO2's 200/400Gb tapes will be OK and not that expensive, drive ~£200, tapes ~£15-20 each.
 
We are using 5 tapes a day and find that LTO's are pretty good for what we do. Although we manage to avoid do a lot of recovery stuff. Based on the amount of tapes that we purchase we manage to get them for around £15 each.
