Triple slot GPU

Depends on the card, the ones that are exactly 3 slot then yes, the ones that are 3 and a bit slots then no.

Also if what you're putting in that slot is big it can block airflow to the graphics card

My card (XFX 7900 XTX) is basically 4 slots with the mounting bar. I have a sound card in a 5th slot below which does give enough clearance due to the bar and upward blowing fans below it. At east I've had no heat issues yet.
 
It will depend on the board anyway because what little pci-e slots we have these days are scattered all over the place, there is no set layout.
