Thought this was worth posting because it kind of flew under my radar as Plex "just works" for me.
In addition I've only used Plex with iOS 5.0.2 so it's never completely worked as I'd have hoped due to the fact that I could never get it in the top row.
(By being in the top row Plex can display your media right from when you turn on your ATV2)
That's solved with this update - so I'm now totally happy

To get Plex in the top row you need to hide another icon. To do this:
1) Go to Settings -> General -> Parental Controls
2) Turn it on put in 0000 as your password (default or whatever you want)
3) Choose one of the top row things to hide (I chose to hide iTunes Match) and enable everything else (or not, up to you)
Restarting your ATV2 at this point should cause Plex to the be the left most icon and as such the default selection when your ATV2 wakes from sleep.
Now you want to make sure it displays what you want up there. To configure it:
1) Go to Plex -> Settings -> View Settings
2) Scroll down until you see "Top Shelf"
Combined sections means it will display both recently added Movies and TV Shows
Disable that and you can choose what section (TV or Movies) and in turn what filter to use (On Deck or Recently Added)
If you don't know On Deck is a neat filter that just lists the next episodes of TV Shows you should watch based on the last thing you watched and the next unwatched episode.
