Using other objects to prevent ESD

As far as I know, anti static stuff uses metal embedded to bleed off any build up of charge, therefore running a mobo on a mat would probably short it out. I would try to use standoffs if possible.
 
If it's just for the purpose of testing then any normal cardboard would be ok to sit it on or the box in which it came.

Its going to sit there for a while, I dont really want an enclosed case... but there no open air cases to fit an E-ATX Motherboard as far as I know. so might have to make some holes in there... Do i drill all the way through the material im using for risers? (Plexi/Acrylic.)
 
It would depend on how thick the material you are using is, measure the screw thread part of a riser and add a mm or two to that to get your drill depth. Being honest i have always used pre made cases so i might not be the best person to advise on anything other then those although i am sure you will find someone on here that is familiar. I will try and help as much as i can but we might hit a wall where my knowledge is not good enough just so you know :).
