Using two PCI-E x16 slots

If I decide to use both PCI-E x16 slots on my mobo, one for a graphics card and the other for a RAID controller, do I have to enable the other one, or should it be already active?

I have tested a couple of RAID controllers in the slot and nothing is detected. I did try testing a graphics card in the slot earlier on...when I turned the machine on, there was a longish beep and then a blank screen. But i was told this was because the graphics card has to be in the primary PCI-E slot?

BTW, the mobo I am using is a LanParty UT RDX 200.
