Website spec / requirements

I'm thinking about setting up a website and need it to do the following:

Storage and streaming of say 500 x 3.5 minute video files and 2000 audio files

I know how to make basic websites but this is slightly different. I don't want use any sites like Youtube to do this.

How would i go about doing this?

I've been looking into different programs for streaming videos and MP3s and close to buying this at the moment...

I haven't tried it myself, but it sounds good and seems easy enough to skin as well.
