Looking to replace the standard items for something better, is there anything I should go for?

Looking at the Alpine SXE-1025S as an option, have around £60 to spend on four speakers.

Well people recommend only upgrading the fronts, amping them, then just running rears off HU. Then you turn the volume down on the rears, this is due to the rears bring back the soundstage.

You after comps or coaxials aswell?

Was just after 4 coaxials but could do a component setup up front and forget the rears if thats going to be better.

HU is a Sony CDX-S3350, (4x50w) would I still need a amp for comps?

Well looking around there seems to be a lack of separates in that size. So i would say go for a pair of good coaxials for the front and run them off the HU.

Get something like Infinity Reference 4012i's, should be better then stock! Though if you can push the extra £10 then i would say Infinity Kappa 42.5i's.

Have you looked into whether you can fit 13cm (5 1/4") speakers? There is a much bigger range of them and also you can get comp systems a lot easier for the size.
