Will a 5970 fit in a Fractual Define R2?

No sorry.
The 5970 will fit in very few cases. The only two cases it will comfortably fit in, is a Antec 1200 and Corsair 800D.
It may well fit in others but these are the only cases it will fit in that we use for our builds.

and it's even fairly snug in my antec 1200..

They really are BIG cards, plus due to the sound proofing on the fractal i imagine there will be heat issues as the sound reduction material inherently keeps heat in

Yes, in that price range it is almost a shame to keep that extra twenty note in your wallet. 
Magick-You may as well go for the 800d its not that more expensive
I'm not sure if i'm that bothered about the swappable drive bay. And the 700d has no window which means it can be soundproofed better.
Is that case as felxible if you later add watercooling?
+1 the ATCS 840 is AMAZING!!! The 5970 maybe a squeeze but do you need a 5970? a 5870 not do the job?
Yes watercooling can be added space for 2 rads etc and on my mates his 5870 and 2 radiators are close but he also has lots of hdds and multiple dvd drives so mounting a resvoir and pump is a lot harder as the hdd bays and dvd drive bays are the best places for a res and pump....pump at bottom of hdd bay and res in a dvd drive bay.
The 800d would be better though as its HUGE!!! but i just dont like the styling...
