I know there are tons of WHS threads so I'm sorry in advance, but can't find an answer to this question.

I'm tempted to move from a basic Linux Samba box, to running WHS for the simple peice of mind it brings with automated backups of PC's in the house. What I'm wondering though is, do backups have to be done every day at a specific time, or will the homeserver make backups as long as those PC's are turned on?

All configurable (e.g. what PC's at what times) through the WHS back up settings.

Also it doens't do full back ups every time it will just do an incremental back up.

I have mine backing up my HTPC,gaming rig and netbook. Sometimes they are switched on at that time and get backed up other times they dont' and it doens't cause problems.

Also if your M.b support Wake On Lan then if I rememebr correctly that can be used as well.

Backups will be done between the time frame you specify when the machines are turned up.
You can set WHS to wake your pc to do a backup or there are addins that will manage this for you. (Autoexit)
If you don't allow WHS to turn on your machine it won't back it up and if it hasn't backed up a machine for a couple of days it will warn you that the machine needs backing up.
 
The WHS backup proccess will also wake a machine from Stanby or Hibernate. I hvae my PC and my laptop hibernate, and my HTPC sleep, and they all wake up between 2 and 7am and backup.
