Putting together a new build, how is this motherboard for the price? Good overclocker?

Also read that it has mounting holes for S775 coolers? Is this correct?

Yes, all X58 mobos are good. I have the P6T's little brother (P6T-SE) and even that pushes my C0/C1 stepping 920 past 4GHz with complete stability. The favourite mobo's on this forum do seem to be Gigabyte's, however I like Asus boards.

I was about to ask the same thing, I noticed that the P6T is slightly cheaper this week. What're the main difference between the P6T and the P6T deluxe? The P6T listing doesn't have specs, so I can't compare...

Serial Attached SCSI.

I don't think it's the only difference. If you notice the P6T Deluxe v2 has better heat sinks, better on-board audio chip set to name a few.

The P6T Deluxe does indeed have SAS ports, but the P6T Deluxe V2 (which is what seems to be available everywhere now) is almost the same board with the SAS removed. I'm not entirely sure why but I do recall reading about several issues a while ago. I believe Asus just decided to remove the controller altogether because of these issues. The V2 also doesn't have the 775 mounting holes.

For comparison of the Asus P6T SE, P6T & P6T Deluxe V2, see Here

The P6T has a nice range of features I think...
You would've expected Asus' own site to have the correct alignment of features, but noooo. :)

From the specs, they mention that the Deluxe v2 doesn't support Tri-SLI, only 2 Way and Quad, what's that all about?

Hold on, you can fit a S775 heatsink to this board? Will such a heatsink adequately cover an i7 CPU? I'm thinking AC7Pro here.

It's due to the placement of the PCI-E slots. From what I read it'd be technically possible with single slot cards, but of course I can't say for sure.

The P6T and P6T SE support 775 heatsinks. The Deluxe and Deluxe V2 do not. The AC7Pro might be a bit of a push, as i7s run hot. I guess if you bought a retail CPU you could always try the AC7 out, then switch to the stock cooler temporarily if it came to it.
