1 Litre OK?

Should cover you unless you have a leak or spillage whilst filling, I always get two as it's useful to keep for when you want to change fluid in a few months
 
1 litre will be enough. My loop is large, has 2x 7970's a 360 res mounted externally so a lot of tubing and an internal 240 rad. Even with all that it uses just under 1 litre.
 
I always get an extra bottle for what I think will do the trick. I have 3 litres in a drawer for example for when I need to do changes and whatnot. Its always better to have more than it is to have less, also fluids come with a typical 3 year shelf life and if your changing every 6 months as they suggest your gonna need some anyway.

I typically have 1-2 DI water for flushing the system and then whatever fluid I need.

As for filling, I just keep filling until the whole thing is full. Ive got a loop with CPU, 2 GPU's and a 360mm RAD (im adding more rads soon!) and that uses less than 1 pack of tubing and for fluids it uses less than 1 litre. I still like to keep more on hand just in case.
 
Halfords de-ionised water available in a 5L container for about £3-4 quid. Its great for flushing out your loop. I used it for flushing out my Alphacool rads prior to installation, the build quality is excellent so there was no debris when I did this.

You can use de-ionised water as a coolant with the appropriate anti corrosion and biocide additives, you just need to be careful that your tubing is chemically inert. Tubing from Primochill doesn't like ethylene glycol (found in antifreeze/car rad fluid) for example, where if you use tygon tubing you can put most chemicals into your loop without issues See here
