I've never posted anything on Twitter before, before I do I was wondering can people see my email if I post a reply on Twitter?

If your tweets are protected, no-one can see them except your 'accepted' followers.

However, if you tweet someone who is not on your followers, they can see that message and reply to you

It was a reply to something that's been posted that I completely disagree with but I think I'll just leave it and get on with my life.
