I still don't get the second nun's reply.

The joke suggests a few images. Since you're focused on the reply, then you can take that:

Nun 2 wore away the soap. Nun 1 can't find it. It wore away pretty quick.

If you take the whole joke as playful banter:

'OOOOH, WHERE is the SOAP?'
'Yes, it does [feel rather nice], doesn't it!' *proceeds to wear away the soap on nun 1*

Alternatively they are both using it and the words don't matter beyond hinting at the scene.

Some versions of the joke use 'Rather!' or 'Yes, I rather would!' as the reply - english.stackexchange.com is your friend. :p

2. Wear (to put on clothes)
3. Wear (to degrade something by rubbing it / friction. I.e. it wears out the soap).
