I need to separate downstairs from upstairs as I am freezing my nuts off but the place has gables that are not well insulated. I figured I could string some sort of a blanket across the divide and head just upstairs for a bit.
Good idea? Bad idea? Either way i saw these at an airbnb and they struck me as very useful. Especially for green screening which I occasionally do.

Also if someone can give me a quick and dirty way of plugging this hole that would be great.

If you could insulate the thing you're suspending from the ceiling, then it might stop some of the heat rising. But if it's just going to be a blanket, I think the heat will just continue to rise.

Trim the hole (make it a rectangle or circle). Put in a plank behind the plasterboard and glue it to the plasterboard. Cut a new piece of plasterboard to fit the hole. Glue new plasterboard to plank. Plaster and paint.

Or perhaps you could make a virtue of the hole and install an access panel. Just find one of the right side and trim the hole to fit.

Where is that hole and whats it need fixing for, cold draft?

Seeing as you have a gaping hole with a space behind I would be tempted to stuff a load of rock wool in there.

Either then similar to what quartz has said, or if its somewhere you really don't care about the appearance you can just grip fill something else on the top, plasterboard, some osb, some ply (ie anything you have lying around or can easily source.
I tend now to grip fill in patches as opposed to putting something behind them. Pretty easy and as tough as the plasterboard. Avoids that annoying piece of wood you can hit later on at some point.

The blanket's going to go all the way up to the ceiling; partitioning the room.

I'm just amazed at how taut the wire is. Assume there's a ratchet or something?
