Chances are they have different pharmacokinetics based on the vehicle (tablet). They're probably absorbed at different rates / in different places in the body. Just off the top of my head I'd say the "nasal" directed ones would more likely be absorbed nearer the mouth, whereas the full body flu tablets would be absorbed in the intestines to allow a more systemic distribution.
I'm tired this may be rubbish but it makes sense to me
And you'll probably find the more expensive painkillers also include ingredients like caffeine which aid the absorption / action of the painkiller itself, giving you quicker relief.
