basically, when you save a file it's saved into loads of small chunks, sometimes these chunks are just saved at random intervals throughout the disk, so it takes longer to open the file as the hard drive 'looks' for the pieces
defragmentation puts all the pieces together for each file so your file will be all in chunks next to each other.
