Do I Need to "Format" to replace/change MOBO?

A format is not necessary. As long as you make a good job of clearing out all the old drivers then it is no different to a clean install of Windows which has to detect your hardware on it's first boot anyway.

The key is ensuring that Windows will still be able to access the disk drive through the new controller. A nice guide can be found here:
