I just bought an Antec MX-1 Enclosure with a Western Digital Caviar SE16 500GB 5000AAKS and whenever I try to connect to my pc's eSata socket the system completely freezes. USB 2 works but runs very slow. The mboard is an Asus P5N-e Sli and I'm running Vista 32bit Home Premium. Anyone else had this sort of problem? Or any idea how to fix this?

Does your motherboard use the JMicron SATA controller? If so you have to switch it to RAID mode rather than IDE. Also get the latest drivers from the JMircron site, rather than the ASUS site.
Do you try and Hot plug when Windows is already booted up, or do you have it plugged in at Boot time?

Im using a WD MyBook 500Gb drive and I had quite a few issues getting it to work at 1st, but after changing the controller to RAID mode, and getting the latest drivers, it worked. it does however make the boot process longer by about 30 seconds after the Windows Loading bar disappears, my screen just goes black and the computer unresponsive, but after about 30 seconds it works fine.
