Be careful of the weight. 2ft tanks usually hold between 60 and 80L of water, so add the weight of the tank and some gravel and it's a good 90Kg - which is about the same as a burly bloke.

For comparison, my Sony 19" CRT was only 27Kg.

Previous question of it smelling - shouldn't do if you look after the tank properly. Fit a filter if there isn't one already, and change 15-20% of the water weekly. You will drip water when you change it, so don't stick a 4 gang socket or your xbox PSU near the fish tank like it did...
 
If your floorboards can stand it and the desk is sturdy and flat, then it should probably hold it OK. Mine has been on my desk for a year or two with no problems.

Obviously you need to be careful not to splash water near the sockets, and take care not to splash your computer/display during maintenance. It's nice to take your eyes off the screen every so often to see what the fish are up to!
