No, Vista will only format FAT32 partitions up to 32GB in size (in Disk Management you'll only have options for NTFS or exFAT), as it's not recommended for very large partitions and/or vast numbers of files.
You can have much bigger FAT32 partitions, but you'll need a third-party app to do it - probably the simplest and quickest way is the command-line utility fat32format.
