I have 3 drives in total:
2x Raptor 74gb (RAID 0, 32k) (2 equal sized partitions)
1x Samsung 400gb

I have Vista installed on one partition and XP on the other. Then I have the 400gb drive for my media files.

I think this is the best setup if I wanted to run both XP and Vista?

When I install programs and games, do I need to install them on the partition I'm using, or could I install them in one windows partition and use them in both? or on the 400gb drive and use them on both?

What about programs and games?

Most programs and games make updates to the registry and need access to these settings. Best to install them individually on each partition.

You can make this process easier by using a utility like PCMover, which assists migrating program files (and games?)
