No, just go into disk management, right click on your hdd and select change drive letter.

Do you think I'll have a problem with the cooling of the system ? If so what would you recommend to do or fit ?

Are they easy to fit ? Suppose everything will have to be taken apart inside ?
Just remove the stock cooler and the rear fan, or fit it in the top of the case. I would also remove the Gpu, as it will make fitting the new cooler easier.
I fort the connector end bit, went on like a plate thing at back of the motherboard ?
