I have an ASUS X99-S with a 5820 CPU and 4 x 4 sticks of G.Skill DDR4 2800 F4 C16Q 16GRK.

I use Asus Dual Intelligent processors for overclocking and have 100 x 45 and DRAM frequency of 3100.

I want to install a further 2 x 32Gb sticks.

I use Photoshop and process large numbers of images in Adobe Camera Raw. Other apps include After Affects and Premiere Pro.

The X99-S had a 64Gb limit but this is a quote from a post on Techpowerup

"The X99 platform launched with a max 64 GB, but subsequent micro-code updates (brought about by ASUS/G.Skill working with Intel) increased that ability up to 128GB. This same micro-code should be present in nearly any X99 board's BIOS at this point. If you can run 2666 MHz on 100 BCLK (you could not at launch, or 2800, or 3000), then you should be good to go. When the platform launched, you had 2133 MHz, 2400 MHz, and then 3200 MHz as the only working dividers on 100 BCLK, and the rest required 125 BCLK or greater. Today is a different story with most boards (there are still a few stragglers who have not got proper updates)."

What are my most cost effective options?

You're best going for 4x16GB as your board is quad channel. You can run 6 modules and keep quad channel (I've done similar) but boards get really fussy with that setup.

I'd by two sets of 2x16GB kits over a 4x16GB matched set as you'll save loads and they'll work fine.
