I can't seem to get my head around the Koolance QDCs.

Could someone spec me which pair I'd need for:

a) Attaching directly to waterblock
b) In middle of tube (I guess barb each end)

I'm using XSPC 7/16" ID High Flex Tubing though that could be changed as I have to drain loop to fit the disconnects.

I would swap your tubing as it seems there are none specifically for that size
I have gone for 1/2" ID 3/4" OD fittings here although if you look for fittings with the same title but different size then you should be good

Essentially you need to look for matching male and female. If you look at the manufacturers code on the product page they begin with something like Vl3N or VL4N or something like that. These fittings will work together. Besides that then you are just finding the other opened to suit. You want a G1/4 thread for screwing into the water block and then your best with a compression fitting for the correct size tubing.
I would choose compression over bards for QDC's as they will get moved about and compressions are more secure

Ok for attaching to the water block:
Female with compression
Male with G1/4 thread

And for joining tubing:
Compression Female
Compression Male

Thanks, exactly the advice I was looking for. Never used compression fittings before, current setup is just barbs.

Do I need to make a cleaner (ie straighter) cut on the tubes when using compression vs barbed fittings?
 
You should try and do a straight cut for both really but there is less tubing in the actual fitting on compressions so i would guess it is slightly more important
 
I wouldn't get the versions that screw directly into blocks tbh. They require a little bit of force to release these mechanisms - enough to put too much pressure on a pci-e slot etc.

Imo the g1/4 thread ones are suited for screwing into bulkheads fitted to the case.
